Identification of Fibronectin Adhesins on Candida albicans
نویسنده
چکیده
475 Abstract: The pattern of distribution of fibronectin adhesins on yeast and hyphalform Candida albicans was studied with goldlabelled fibronectin and transmission electron microscopy. The germ-tube parts of the hyphal-form cells were the major binding sites for gold-labelled fibronectin, and the attached yeast-cell parts and yeast-form cell remained unlabelled. Therefore, the walls of the germ tube parts of hyphal-form cells might be sites of important interactions between C. albicans and host tissues containing fibronectin.

The fibronectin adhesin of Candida albicans.
Candida albicans possesses on its cell surface an adhesin which binds the whole viable fungus to subendothelial extracellular matrix and matrix proteins. The adhesin is composed of 75 to 80% carbohydrate and approximately 20 to 25% protein by weight. High-performance liquid chromatography of material eluted from a fibronectin-agarose affinity column demonstrates the presence of three peaks, all...
متن کاملCandida albicans adhesins: Biochemical aspects and virulence.
The recognition of host cells by the pathogenic yeast, Candida albicans, is probably an essential step in the pathogenesis of disease development. The interaction of yeast and hyphal mannoproteins with host cell receptors has been studied by a number of laboratories. C. albicans recognizes a variety of host cells as well as host cell extracellular matrix proteins. This observation is not unexpe...
متن کاملInhibition of adherence and killing of Candida albicans with a 23-Mer peptide (Fn/23) with dual antifungal properties.
Candida albicans adheres to host tissue and then proliferates in order to establish a commensal as well as a pathogenic state. Specific adherence to proteins is provided by several surface adhesins of Candida. Two well-studied proteins, Als1p and Als5p, do not require energy for adherence to occur (dead as well as living cells adhere) and have a multiplier effect of cell-cell aggregation that m...
متن کاملGlobal cell surface conformational shift mediated by a Candida albicans adhesin.
Candida albicans maintains both commensal and pathogenic states in humans. Both states are dependent on cell surface-expressed adhesins, including those of the Als family. Heterologous expression of Als5p at the surface of Saccharomyces cerevisiae results in Als5p-mediated adhesion to various ligands, followed by formation of multicellular aggregates. Following adhesion of one region of the cel...
متن کاملIdentification of Candida albicans genes that induce Saccharomyces cerevisiae cell adhesion and morphogenesis.
Morphogenesis and adhesion to host tissues and medical devices contribute to the virulence of Candida albicans, the most common fungal pathogen isolated from humans. However, identification of molecular mechanisms of C. albicans adhesion and morphogenesis has been impaired by the lack of effective molecular and genetic tools available for this organism. Saccharomyces cerevisiae provides an attr...
